  报告摘要Diaphorase (DI) is an enzyme capable of transferring electrons between NAD(P)H and chemical electron mediators such as benzyl viologen (BV). Biomimetic electron transport chains, comprised of an NADH-dependent or NADPH-dependent DI and a suitable electron mediator, were designed to enhance the electron transfer rates by lowering activation energy barriers. The H2 generation rates via the biomimetic chains increased by approximately five-fold compared to those catalyzed by [NiFe]-hydrogenase only. We demonstrated an in vitro enzymatic pathway that can produce H2 at extremely high rates by splitting water energized by starch in the stoichiometric reaction C6H10O5 + 7H2O -> 12H2 + 6CO2. This pathway of fifteen hyperthermophilic enzymes is comprised of ATP-free starch phosphorylation, an NAD- rather than NADP-based pentose phosphate pathway, a BV-conjugated DI, and a [NiFe]-hydrogenase. The highest reaction rate of 530 mmole H2/L/h (i.e., more than 11 L of H2 per L of bioreactor per h) was accomplished at 80°C. The concept of biomimetic electron transport chains could also be applied to biosystems for artificial photosynthesis. A plausible application scheme of the biomimetic chains for CO2 fixation will be elucidated.   

  报告人简介:Eui-Jin Kim博士分别于2001年、2010年于Sogang University获得理学学士学位和博士学位,并于20103-8月在Sogang University 做博士后研究。20109- 20157月在Sogang University作研究员,20125- 20154月作为项目负责人获得由韩国国家研究基金会(NRF)资助的研究项目,研究叶绿素/细菌叶绿素反应机理合成酶,确定底物特异性和抑制剂相互作用的决定因素,取得了重大进展。20158 月至今,在美国弗吉尼亚理工大学作助理研究员,主要研究领域是利用电子传递链在体外多酶体系的应用来进行高效的氢气生产。金博士在ChemistryJ. Microbiol. Biotechnol,J. Bacterial等杂志上发表了多篇论文,并申请专利10余项。
